We are having the same issue:

Error is:
Destination Folder Does Not Exist!
Rerun the Wizard and Create the SalesCart Web first.

Then another box comes up:
SalesCart Transaction Plug-in
Unable to load documents into web and cannot continue

If we want the TS.asa to be created in c:\sites\site1

On my 'Web Site General Properties' page my
Web Site Path is:
c:\sites\site1

Web Site name is:
sitename

That was it. I appended the Site Name to the root directory. Problem solved. It sounds like you don't have the right folder. I have the development site up and running just fine. I'm attempting to install the transaction processing bridge to connect to authorizenet. When I run the wizard it tells me "Destination folder does not exist."
